Glucovance 1000 mg/5 mg Tablets (30's)

Active Ingredients:

  • Glyburide (Glibenclamide) 5 mg (Sulfonylurea class)
  • Metformin Hydrochloride 1000 mg (Biguanide class)

Therapeutic Class:

  • Antidiabetic agent (Combination of sulfonylurea and biguanide)

Indication:

  • Used to manage type 2 diabetes mellitus in adults.
  • Helps control blood sugar levels when diet and exercise alone are insufficient.
  • Metformin reduces glucose production in the liver and improves insulin sensitivity.
  • Glyburide stimulates the pancreas to release more insulin.

Mechanism of Action:

  1. Glyburide (Sulfonylurea):

    • Stimulates the pancreatic beta cells to release more insulin, helping to lower blood glucose levels.
    • Reduces glucose production in the liver.
  2. Metformin (Biguanide):

    • Reduces hepatic glucose production.
    • Increases insulin sensitivity in peripheral tissues, making the body use insulin more effectively.
    • Slows down intestinal glucose absorption.

Dosage & Administration:

  • Taken once or twice daily, with meals, to reduce the risk of gastrointestinal side effects.
  • Tablets must be swallowed whole; do not crush, split, or chew.
  • The dose is individualized based on blood glucose levels and tolerance.

Contraindications:

  • Severe renal impairment (eGFR < 30 mL/min/1.73m²)
  • Metabolic acidosis (including diabetic ketoacidosis)
  • Hypersensitivity to glyburide, metformin, or any excipients
  • Pregnancy and breastfeeding (unless prescribed by a doctor)

Warnings & Precautions:

  • Regular monitoring of renal function is essential, especially during the initiation of treatment.
  • Risk of lactic acidosis in cases of renal or hepatic impairment.
  • May cause hypoglycemia (especially with glyburide), so monitor blood sugar closely, particularly when combined with other hypoglycemic agents.
  • Alcohol consumption should be minimized, as it increases the risk of lactic acidosis and hypoglycemia.
